| Description: | HIV-1 RT RDDP activity was generally determined by a fixed time assay. Briefly, reaction mixtures contained p51/p66 RT, template-primer, and [3H]-TTP substrate. Aliquots of DMSO solutions containing the inhibitor were added such that the final DMSO concentration was < 2%. Reaction assays were incubated at 37 deg C for 10-20 min and then quenched with sodium pyrophosphate in 10% trichloroacetic acid (TCA). The samples were then filtered on Whatman 934-AH glass fiber filters and washed with 10% TCA and ethanol, and the radioactivity determined by liquid scintillation spectrometry. |
