| Measurement of hBuChE Activity |
Description: | The inhibitory activity of the compounds towards BuChE was determined following the method of Ellman using BuChE from human serum and butyrylthiocholine chloride as a substrate. The reaction took place in a phosphate buffer solution containing BuChE and 5,5-dithiobis-2-nitrobenzoic acid (DTNB) which produces the yellow anion 5-thio-2-nitrobenzoic acid. Inhibition curves were made by incubating the reaction mixture with the different compounds for 15 min; a sample without any compound was always present to determine the 100% of enzymatic activity. After the 15 min incubation period, the production of color, as an indication of enzymatic activity, was evaluated by measuring absorbance at 412 nm in a spectrophotometer plate reader (iEMS Reader MF, Labsystems). |
