| Description: | All assays against Arb93A using the PACE method were carried out as described by[Carapito et al.[Carapito et al., J. Biol. Chem., 284:12285] using inhibitor concentrations of 50 and 100 μM. Assays against Arb93A using 4 were carried out in triplicate at 40°C using a continuous assay procedure where reactions were initiated by the addition of enzyme. Assays were conducted in buffer (20 mM Tris, 200 mM NaCl, pH 8.3) For the extent of 4-nitrophenol release this was determined using a Cary 4000 at 400 nm. Assays usedenzyme at a concentration of 0.5-10 nM with the appropriate substrate concentration. |
