| Description: | To assess the effect of inhibitors, RT RNH activity was measured using a FRET-based microplate fluorescence assay. Briefly, the RNA-DNA hybrid duplex was added to individual wells of a 96-well microplate containing test compound. Reactions were initiated by the addition of HIV-1 RT and allowed to proceed at 37 deg C for 30 min. Reactions were quenched by the addition of EDTA (0.5 M). Fluorescence intensity was assessed using an excitation wavelength of 490 nm and an emission wavelength of 528 nm, with cutoff filter set to 515 nm. |
