| Description: | The reporter assay for the stable hGPR40-Gal4-Elk1/5xGal4-luc+-expressing CHO cells was first carried out in the serum-free medium in the presence of test ligand for 5h. The medium was then removed and replaced with 1:1 mixture of LucPlusTM (PerkinElmer Life Sciences) and Dulbecco phosphate buffered saline containing 1 mM CaCl2 and 1 mM MgCl2. Plates were incubated in the dark at room temperature for 10 min before luciferase activity was determined using a TopCountTM microplate scintillation counter (PerkinElmer Life Sciences) at a count time of 3 s/well. |
