| Description: | This assay adopted PARP1, TNKS1, TNKS2, PARP7 and PARP14 chemiluminescence detection kits (BPS, Cat No. 80551/80552/80573/80578/79729/80568) for enzymatic biochemical assay of PARP. The specific scheme was as follows: the 1× histone mixture was added to a 96-well plate at 50 μL per well, and the plate was incubated overnight at 4° C. The next day, after the plate was washed with PBST, 200 μL of blocking buffer per well was added, and the plate was incubated for 90 min. After the plate was washed with PBST again, 5 μL of inhibitor, 20 μL of 1×PARP buffer and 25 μL of streptavidin-HRP were added to each well, and the plate was incubated at room temperature for 30 min. After the plate was washed with PBST, 100 μL of Elisa ECL substrate A and B mixture was added to each well, chemiluminescence value was immediately read using a plate reader, and IC50 values were calculated. |
