| Description: | Full length human CDC7 protein co-expressed with DBF4 was purchased from SignalChem (China). CDC7 kinase activity was determined with PDKtide (SignalChem) as a substrate and by measuring ADP production using the ADP-Glo Kinase Assay kit (Promega) following the manufacturers instructions. The kinase reaction was performed using the following conditions: Buffer: 40 mM Tris pH 7.5, 20 mM MgCl2, 0.1 mg/ml BSA and 50 uM DTT. Final reaction mix contained 0.1 nM CDC7/DBF4, 1 uM ATP and 10 uM PDKtide. The kinase reaction time was 4 h. The ADP-Glo signal was measured using an EnVision plate reader (PerkinELmer). |
