Description:  Fluorescence polarization assay (FP assay) was used to detect the affinity of compounds for human STING proteins. There were a certain amount of fluorescein-labeled c-di-GMP and different concentrations of test compounds in the reaction system. When the C-terminal protein of recombinant human STING was added, two small molecules competitively combined with the protein. The rotation of the fluorescein-labeled c-di-GMP in the bound phase is relatively slow in the liquid phase, and the degree of fluorescence polarization detected is relatively high. The degree of fluorescence polarization is inversely proportional to the concentration and affinity of the compound to be tested. By detecting the size of the polarized light in the reaction system, we can accurately know the affinity of the compound to be tested for human STING.
